I was able to resolve Delimiter (added last row in master file) but not sure how to add Header = 'Yes' option in master file, any help would be much appreciated.

A master file is to define how the data is organized where the acx file is to define how/where to access the data. And AFAIK, HEADER=YES is telling FOCUS how to access the data by skipping the first row since it's not a data row.
If FOCUS create .acx and .mas it's for a good reason, you should not attempt to bypass that
